AAV Recombinase
The Cre-LoxP site-specific recombination system is a powerful tool for genomic editing, enabling precise deletion, insertion, translocation, and inversion of DNA in both eukaryotic and prokaryotic organisms. This system is composed of two main elements: the Cre recombinase enzyme and specific DNA sequences known as LoxP sites. Cre recombinase, a 38 kDa protein encoded by bacteriophage P1, interacts with LoxP sites by binding to two 13 base pair recombinase binding elements (RBEs). These RBEs are arranged as inverted repeats surrounding an 8 base pair asymmetric region responsible for strand exchange, together forming the LoxP site. The orientation of the LoxP site is determined by this central 8bp sequence.
